MUC-1 antigen is a mammary-type apomucin, a high molecular weight transmembrane glycoprotein, of which the extracellular domain is formed by a repeating 20 amino acid sequence (in tandem) with a high content of serine and threonine on which are O-linked carbohydrate chains. MUC-1 synthesis and secretion are features of glandular epithelial tissues; MUC-1 is overexpressed in lactating breast and in breast, ovary, lung, and prostate malignancies.

Irinotecan, a semisynthetic derivative of camptothecin, inhibits topoisomerase I activity by stabilizing the cleavable complex between topoisomerase I and DNA, resulting in DNA breaks that inhibit DNA replication and trigger apoptotic cell death.

Human autologous peripheral blood lymphocytes (PBLs) transduced with an anti-p53 T cell receptor gene with potential antineoplastic activity. PBLs are harvested from a patient and pulsed with a retroviral vector that encodes the T-cell receptor gene specific for a mutated form of p53. The transduced PBLs are then expanded in culture. When reintroduced to the patient, these modified PBLs express the anti-p53 T cell receptor which binds to mutant p53-overexpressing tumor cells; PBL-mediated tumor growth inhibition may follow. Many tumor cell types overexpress mutant p53 proteins, which are associated with the loss of apoptosis regulation and abnormal cell proliferation.

Human autologous peripheral blood lymphocytes (PBLs) transduced with a retroviral vector encoding an anti-NY-ESO-1 T cell receptor (TCR) gene with potential antineoplastic activity. PBLs are harvested from a patient and pulsed with a retroviral vector that encodes the T-cell receptor gene specific for NY-ESO-1. After expansion ex vivo, the transduced autologous PBLs expressing anti-NY-ESO-1 TCR are reintroduced into the patient and bind to NY-ESO-1-overexpressing tumor cells. This may stimulate the immune system to mount a cytotoxic T-lymphocyte response (CTL) against NY-ESO-1 expressing tumor cells. NY-ESO-1, a tumor associated antigen (TAA), is found in normal testis and on the surface of various tumor cell types.

MART-1 (melanoma antigen recognized by T cells 1), also known as Melan-A, is a melanocyte lineage-specific transmembrane protein.

After expansion ex vivo, the transduced autologous PBLs, expressing this specific TCR, are reintroduced into the patient and bind to melanoma cells expressing the gp100 protein, which may result in specific cytotoxic T-lymphocyte (CTL) killing of gp100-expressing melanoma cells. Gp100 is a melanocyte lineage-specific antigen overexpressed in melanomas.
